چکیده

Cell mechanical characterization has recently approached the throughput of conventional flow cytometers. However, this very sensitive, label-free approach still lacks the specificity of molecular markers. Here we combine real-time 1D-imaging fluorescence and deformability cytometry (RT-FDC) to merge the two worlds in one instrument — opening many new research avenues. We demonstrate its utility using sub-cellular fluorescence localization to identify mitotic cells and test for their mechanical changes in an RNAi screen. peer-reviewed) is the author/funder. Microfluidic electroporative flow cytometry for studying single-cell biomechanics.

Biomechanical properties of cells yield important information on the disease state of cells such as transformation and metastasis. Screening of cells based on their biomechanical properties provides rapid tools for label-free diagnosis and staging of cancers. However, existent single-cell techniques for measuring biomechanical properties suffer from low throughput (<1 cell/min). This prevents t...

P-6: Flow Cytometry: Assessment of Protamine Deficiency by CMA3 Staining and Apoptotic Bodies

Background: Chromomycin A3 (CMA3) staining, with the slide method or fluorescence microscopy, is widely used for indirect assessment of protamine deficiency in a semen sample. Flow cytometry is the most suitable tool which to improve assessment accuracy, both in terms of statistical analysis and to prevent observer variation. This study provides a simple procedure to account for merocyanine 540...

A mechanical biomarker of cell state in medicine.

The mechanical properties of cells have been shown to be useful markers of cell state by the biophysics community. Here, I highlight clinical and research problems that this label-free, potentially inexpensive cellular biomarker can address and discuss technical challenges to realize automated instruments to achieve robust and high-throughput mechanical measurements. Important features found in...

Deformability-based flow cytometry.

BACKGROUND Elasticity of cells is determined by their cytoskeleton. Changes in cellular function are reflected in the amount of cytoskeletal proteins and their associated networks. Drastic examples are diseases such as cancer, in which the altered cytoskeleton is even diagnostic. This connection between cellular function and cytoskeletal mechanical properties suggests using the deformability of...

A FRET-based ratiometric redox probe for detecting oxidative stress by confocal microscopy, FLIM and flow cytometry.

Understanding the role of oxidative stress in disease requires real time monitoring of redox status within a cell. We report a FRET-based, ratiometric redox probe which can be applied to monitor cellular oxidative capacity using three different modalities – confocal microscopy, fluorescence lifetime imaging and flow cytometry.
